[摘要]
目的运用基于1H-NMR和UPLC的植物代谢组学技术对生远志、蜜远志及甘草制远志进行代谢组学分析。方法首先采用基于1H-NMR及UPLC代谢组学技术对生远志及两种炮制品进行分析,指认初级代谢产物21种,次级代谢产物8种,并采用UPLC技术测定三者中细叶远志皂苷的量。结果生远志与蜜远志、甘草制远志化学成分明显不同|与蜜远志相比,生远志与甘草制远志化学成分更加接近|三者中部分氨基酸、有机酸及糖类等的量变化较大|与生远志相比,蜜远志中总皂苷量几乎不变,甘草制远志则有所上升|糖酯类化合物3,6′-二芥子酰基糖酯在甘草制远志中其量降低,蜜远志中最低,同时部分次级代谢产物的量也有所不同。结论从代谢组学整体角度来看,生远志及其蜜制、甘草制品化学成分差异明显,说明炮制可改变药性,导致其功效可能各有不同。

[Abstract]
Objective Metabolomic analyses of crude, honey-stir-baked (HSB-), and liquorice-stir-boiled (LSB-) Polygalae R. (PR) by using plant metabolomic technology based on 1H-NMR and UPLC. Methods First, a series of metabolites were identi directly from 1H-NMR spectra and UPLC chromatograms of the above three samples, twenty-one primary and eight secom metabolites were assigned. Then, the content of tenuifolin in the three samples were determined by UPLC. Results The rep showed that the chemical constituents in crude PR were obviously different from the processed two samples. Compared, HSB-PR, the chemical constituents in another two samples were similar. The contents of amino acids, organic acid, and carbohydr changed greatly among the three samples. Compared with crude PR, the content of total saponins in HSB-PR was almost the s but increased in LSB-PR. Meanwhile, the content of 3, 6'-disinapoylsucrose ester in crude PR was the highest, LSB-PR was second, and the lowest was in HSB-PR. At the same time, the contents of some secondary metabolites were different. Conclu From the metabonomic perspective, chemical constituents in crude, HSB-, and LSB-PR are very different, which shows that processing may change the medicinal properties so as to lead the various pharmacological activities.
