目的观察地榆总皂苷对3种造血细胞Baf3/Mpl、UT-7和NSF-60的促增殖效应。方法分别培养依赖血小板生成素(TPO)、红细胞生成素(EPO)和粒细胞集落生长因子(G-CSF)生长的Baf3/Mpl、UT-7和NSF-60细胞,在加或不加细胞因子条件下,用不同质量浓度地榆总皂苷处理细胞72 h后,采用MTT法检测3种细胞的增殖;采用RT-PCR法检测3种细胞因子受体表达水平的变化。结果地榆总皂苷单独处理细胞72 h,明显促进Baf3/Mpl细胞和NSF-60细胞增殖,但不影响UT-7细胞生长;在TPO和G-CSF存在条件下,地榆总皂苷能分别增强两种细胞因子的促细胞增殖作用。此外,地榆总皂苷能明显增强Baf3/Mpl细胞TPO受体表达,对NSF-60细胞G-CSF受体表达没有影响。结论地榆总皂苷具有单独或协同细胞因子的促造血细胞增殖作用,其促增殖作用与上调TPO受体的表达有关。

ObjectiveTo investigate the proliferative effects of total saponins from Sanguisorba officinalis (TSS) on three hematopoietic cell lines.Methods Three kinds of hematopoietic cells (Baf3/Mpl, UT-7, and NSF-60) expressing cytokines such as thrombopoietin (TPO), granulocyte colony-stimulating factor (G-CSF), and erythropoietin (EPO), were cultured with or without cytokines. After 72 h the TSS at different concentration was added, cell proliferation capacity was estimated by MTT, and expression level of cytokine receptors was measured by RT-PCR. Results TSS promoted the proliferation of Baf3/Mpl and NSF-60 cells but had no effect on growth of UT-7 cells. With TPO and G-CSF, TSS could enhance the effect of promoting cell proliferation of the two cytokines, respectively. TSS could increase the expression of TPO receptor in Baf3/Mpl cells, but had no effect on the expression of G-CSF receptor in NSF-60 cells. Conclusion TSS could promote the proliferation of hematopoietic cells alone or in combination with cytokines. In Baf3/Mpl cells, the proliferative effect is correlated with the up-regulation of TPO receptor.
