目的 观察罗布麻叶总黄酮的降压作用,探讨其作用机制。方法 50只雄性SD大鼠随机分为对照组,模型组,罗布麻叶总黄酮高、低剂量(32、8 mg/kg)组和罗布麻降压片(0.4 g/kg)阳性对照组。除对照组外,其他各组在喂饲高脂高盐饲料诱导大鼠高血压模型的同时,每天ig给予不同剂量的药物,并运用无创血压测量仪监测大鼠血压的变化。造模6周后,利用免疫组化法检测血管组织转化生长因子-β1(TGF-β1)蛋白表达,Western blotting法检测C-反应蛋白(CRP)表达,分光光度法检测血清中高密度脂蛋白-胆固醇(HDL-C)、低密度脂蛋白-胆固醇(LDL-C)、总胆固醇(TC)、三酰甘油(TG)水平。结果 高脂高盐饲料能显著升高大鼠血压,改变血管组织中CRP、TGF-β1的表达。与模型组比较,罗布麻叶总黄酮在高剂量时能降低血压,抑制血管组织中CRP、TGF-β1的表达,提高血清中HDL-C的水平,降低LDL-C、TC、TG的水平。结论 罗布麻叶总黄酮具有显著的降压作用,其机制与调节血脂代谢、抑制CRP和TGF-β1因子的表达有关。

[Abstract]
Objective To study the antihypertensive effect of the extracts from total flavonoids in Apocynum venetum leaves (TFA), and eluciate their mechanisms. Methods Fifty male SD rats were randomly divided into five groups: Normal, model, TFA high- and low-dose (32 and 8 mg/kg), and positive control (Luobuma Tablets, 4g/kg) groups. Except normal group, all other groups were fed with a specially-made high-salt, high-fat diet, and 5% salt water, in addition animals in drug groups were respectively treated with different doses of TFA by ig administration and the blood pressure of rats was observed through a non-invasive blood pressure measurement system. After feeding for six weeks, the protein expression levels of TGF-β1 and CRP in vascular tissue were determined and observed by immunohistochemistry and Western-blotting methods, the levels of HDL-C, LDL-C, TC, and TG in serum were detected by spectrophotometry. Results TFA could significantly inhibit the damage induced by high-salt, high-fat diet, and lower blood pressure via adjusting blood-lipoids metabolism and controlling CRP and TGF-β1 expression, increasing HDL-C level, and decreasing LDL-C, TC, and TG levels in serum as comparing with the model group. Conclusion The antihypertensive effect of TFA may be related to regulating blood lipids and inhibiting the expression of CRP and TGF-β1.
