[摘要]
目的 探讨柞蚕雄蛾提取液对放疗后荷瘤大鼠肿瘤组织缺氧诱导因子-1α(HIF-1α)表达的影响。方法 制备W256荷瘤模型,常规放疗后,柞蚕雄蛾提取液16.53 mg/kg连续ig给药10 d。采用免疫组化染色检测肿瘤组织中HIF-1α和血管内皮生长因子(VEGF)的表达;原位杂交法检测肿瘤组织中HIF-1α mRNA表达。结果 免疫组化结果显示,与模型组比较,放疗组肿瘤组织HIF-1α和VEGF蛋白表达有增强趋势(P>0.05);放疗联合柞蚕雄蛾提取液(联合组)和柞蚕雄蛾提取液组肿瘤组织HIF-1α和VEGF的表达均显著减弱(P<0.05),且HIF-1α与VEGF呈明显正相关(r=0.649,P<0.01)。原位杂交结果显示,放疗组较模型组HIF-1α mRNA表达无统计学意义(P>0.05);联合组及柞蚕雄蛾提取液组肿瘤组织HIF-1α mRNA表达均降低(P<0.05)。结论 柞蚕雄蛾提取液对放疗后及未实施放疗的肿瘤组织HIF-1α的表达有下调作用。

Objective To investigate the effect of the extract from male zooid of Ahtheraea pernyi on the expression of hypoxia-inducible factor-1α (HIF-1α) in tumor-bearing rats after radiotherapy. Methods W256 tumor model was established by continuous ig administration of the extract from the male zooid of A. pernyi (16.53 mg/kg) for 10 d after conventional radiotherapy. To observe the expression of HIF-1α and vascular endothelial growth factor (VEGF) in tumor tissue by immunohistochemical staining and the HIF-1α mRNA expression by in situ hybridization (ISH). Results Compared with the model group, immunohistochemistry results showed the expression of HIF-1α and VEGF increased with tendency (P>0.05) in tumor tissue of the rats in radiotherapy group, and the HIF-1α and VEGF expression in the combination group and the extract from the male zooid of A. pernyi group decreased significantly (P<0.05). HIF-1α and VEGF were correlation positively (r = 0.649, P<0.01). ISH results revealed the expression of HIF-1α mRNA was no statistical significance (P>0.05) in radiotherapy group compared with the model group. However, the HIF-1α mRNA expression in tumor tissue was lower in both the combined group and the extract from male zooid of A. pernyi group (P<0.05). Conclusion The extract from male zooid of A. pernyi could down-regulate the HIF-1α expression in tumor tissue after radiotherapy or without radiotherapy.
