目的 研究黄芪醇提取物对糖尿病大鼠冠状小动脉平滑肌张力的影响。方法 通过 ip 小剂量链脲佐菌素 (STZ) 及高脂饮食的方法诱导2型糖尿病大鼠模型,应用离体小动脉灌流技术研究去除血管内皮后,累积浓度黄芪醇提取液对正常大鼠和糖尿病大鼠冠状小动脉平滑肌基础张力的影响;观察黄芪醇提取物对苯肾上腺素 (PE) 或 KCl 预收缩的微血管环的作用。结果 TZ 和高脂饮食可致 Wistar 大鼠形成2型糖尿病,模型大鼠体质量与对照组比较明显减少,模型组大鼠与对照组大鼠相比血清胆固醇 (TC)、甘油三酯 (TG) 和游离脂肪酸 (FFA) 差异具有统计学意义 (P<0.05),糖化血红蛋白 (HbAlc) 和空腹血糖值明显增高,并出现高脂血症 (P<0.05)。黄芪醇提取物对糖尿病大鼠冠状小动脉环有舒张作用;对 PE 和 KCl 预收缩的去内皮冠状小动脉产生浓度依赖性舒张作用。结论 黄芪醇提取物对糖尿病大鼠冠状小动脉有舒张作用。

[Abstract]
Objective To investigate the effect of Astragali Radix(AR) alcohol extract on smooth muscle tension in coronary small artery of diabetic rats and the underlying mechanism. Methods Rat′s high glucose model was established by ip injection of Streptozocin (STZ) and feeding with high fat diet. Videomicroscopy technique was used to determine the effect of several drugs on the vasomotion of coronary small arteries. Results Type 2 diabetic models of Wistar rats were induced by STZ and high fat diet. The change of body weight in diabetic rats obviously increased than that in nondiabetic rats. TC, TG, and FFA showed statistically significant difference (P<0.05), HbAlc and the fasting blood sugar of diabetic rats obviously increased, and the mod els of hyperglycemic rats were made (P<005). AR Extract could dilate the coronary small artery rings of diabetic rats. Moreover, it caused a concentrationdependent relaxation while artery rings were preconstricted with PE or KCl. Conclusion We observe the effects of contraction and dilation dysfunction of coronary small arteries in diabetic rats induced by STZ and high fat diet. The results indicate that AR can dilate the coronary arteries of diabetic rats.
