[摘要]
目的分析血水草生物碱(ECA)致钉螺肝脏蛋白质表达的变化。方法10mg/LECA浸泡钉螺36h后,解剖活钉螺并收集肝脏,双向电泳分离ECA组和清水对照组钉螺肝脏蛋白质,ImageMaster2D5.0软件分析图谱,筛选差异蛋白质,并用MALDI-TOF-TOF质谱进行鉴定。结果ECA组和清水对照组分别检出(354±110)、(311±12)个蛋白点;质谱鉴定获得假定蛋白、ENSANGP00000022175蛋白、醛脱氢酶、未知蛋白、ENSAN-GP00000027067蛋白、类似锚蛋白3的1亚型4亚亚型蛋白、相似A型肝炎病毒短型细胞受体1的蛋白、线粒体异柠檬酸脱氢酶2样蛋白、类似角蛋白的蛋白、角蛋白10、角蛋白1共11个蛋白质,这些蛋白质均为ECA处理后表达上调的蛋白质。结论ECA可引起钉螺肝脏蛋白质表达发生改变。

[Abstract]
Objective To analyze the influence of Eomecon chionanthe alkaloids(ECA) on differential expression of liver proteins from Oncomlania hupensis.Methods O.hupensis was immersed in 10 mg/LECA or clean water for 36h and livers were isolated from live snails.Total liver proteins were extracted and separated by two-dimensional gel electrophoresis,differentially expressedproteins between ECA group and clean water group were selected and analyzed by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI TOF-MS) and tandem mass spectrometry sequencing of tryptic peptides.Results There were (354±110) and (311±12)spots observed in ECA group and in water group,respectively. Eleven differential expressed proteins were identified and all were up-regulated in ECA group.The eleven identified proteins were hypothetical protein,EN SA N GP00000022175,class I aldehyde dehydrogenase,unknown protein,EN SAN GP00000027067,protein similar to ankyrin 3 isoform 1 isoform 4,protein similar to hepatitis A virus cellular receptor 1 short form,mitochondrial isocitrate dehydrogenase 2-like,protein similar to keratin,keratin-10,and keratin-1,respectively. Conclusion Expression of liver proteins from O.hupensis could be changed when immerse in ECA.
