[摘要]
目的对不同生长时期的环草石斛试管苗进行生长动态及分级标准的研究,为试管苗各生长发育时期合理调控技术措施的确定提供科学依据。方法采用常规方法,测定不同生长时期试管苗的生长指标、多糖及蛋白质的量,确定试管苗出瓶时间及分级标准。结果播种生长12个月的时间内,5~8个月是环草石斛试管苗苗高和茎粗增长最快的时期;
7~8个月时的试管苗生物量积累速率最高。环草石斛试管苗根与茎叶之比(R/T)相对较小,比值小于1。生长8个月时的试管苗多糖的量和可溶性蛋白质的量较高,7~8个月时的试管苗生长代谢旺盛。从干物质积累动态、多糖及蛋白质的量变化、试管苗活力和生产成本等方面综合考虑,环草石斛生长8~10个月的试管苗出瓶移栽较为合适。试管苗出瓶后分级再利用能够切实保证移栽成活率。以苗高、茎粗和根数作为分级指标,环草石斛试管苗可以分为3个等级。结论环草石斛的种子质量对试管苗的生长影响较大,生长8~10个月的环草石斛试管苗出瓶移栽较为合适,以苗高、茎粗和根数作为分级指标,试管苗可以分为3个等级加以利用。

[Abstract]
Objective To study the growth dynamic and standard degree of Dendrobium loddigesii test-tube seedling in various developing periods.It would provide the scientific theory for the realization large scale production of the test-tube seedling finally.Methods Indexes,such as growth,development,polysaccharide content,and soluble protein contents,were determined,and indexes of standard degree of D.loddigesii test-tube seedling were obtained by general ways.Results In a year after sowing,the test-tube seedling of 5—8 months-old was the fastest growing period in height and stem thickness;the test-tube seedling biomass accumulation rate of 7—8 months old was the highest.The test-tube seedling of R/T1.Eight months-old had higher polysaccharide content and soluble protein contents.The test-tube seedling of 8—10 months-old transplanted better on thinking of dry matter accumulation dynamics,polysaccharide content,soluble protein contents variation,seedling activity,and production cost.Classification and reuse after bottle may improve the transplanting survival ratio.Seedlings of D.loddigesii may be divided into three grades by clustering statistic analysis on classification indexes,such as height,stem thickness,and root number.Conclusion The test-tube seedlings of 8—10 months-old transplant better.Seedlings of D.loddigesii may be divided into three grades by clustering statistic analysis on classification indexes,such as height,stem thickness,and root number.
