目的研究三七总皂苷(PNS)对体外培养大鼠心肌细胞肥大及在体大鼠心肌肥大的影响。方法体外培养大鼠心肌细胞,用血管紧张素(Ang)刺激制备心肌细胞肥大模型,分别采用Lowry法、3H-亮氨酸掺入法和激光共聚焦显微镜测定细胞内蛋白质的量、蛋白质合成速率和游离钙荧光强度。ip去甲肾上腺素法诱导大鼠心肌肥大,计算心脏指数和左心室指数。结果与模型组相比,0.05、0.10、0.15g/LPNS可显著降低细胞内蛋白质的量和3H-亮氨酸掺入量,减弱细胞内Ca2+荧光信号。25、50、75mg/kgPNS可显著降低心肌肥大大鼠左心室指数。结论PNS对Ang诱导的大鼠心肌细胞肥大和去甲肾上腺素诱导的在体大鼠心肌肥大均有明显的抑制作用。

[Abstract]
Objective To study the effects of Panax notoginseng saponins (PNS) on myocardial hypertrophy in vitro and in vivo in rats. Methods Cardiomyocytes of rats were isolated with mechanical dissection, enzyme digestion, and cultured in vitro. Cardiomyocyte hypertrophy was induced by Angiotensin Ⅱ (Ang Ⅱ). The protein content, protein synthesis rate, and free calcium concentration were determined by Lowry's method, 3H-leucine ( 3H-Leu) incroporation method, and laser scanning confocal microscope, respectively. Myocardial hypertrophy in rats was induced by ip norepinephrine. The heart and left ventricular were weighed and their heart indexes were calculated. Results PNS (0.05, 0.10, and 0.15 g/L) significantly decreased the protein content, 3H-Leu incorporation rate, and Ca 2+ fluorescence intensity compared with the model group. PNS (25, 50, and 75 mg/kg) could markedly reduce the left ventricular index of myocardial hypertrophy rats. Conclusion PNS could effectually prevent myocardial hypertrophy induced by Ang Ⅱ and norepinephrine in vitro and in vivo in rats
