[摘要]
目的建立了青海地区唐古特大黄药材的HPLC指纹图谱。方法采用反相高效液相色谱法,乙腈-水(0.04%的磷酸)为流动相,梯度洗脱,体积流量1.0 mL/min,检测波长为280 nm,柱温为40℃。结果精密度、重现性、稳定性试验中共有峰面积和保留时间的RSD均小于5%。青海不同采集地唐古特大黄的平均相似度为0.925。结论该方法简便、实用、可靠,可用于以青海果洛地区为主产地不同海拔唐古特大黄药材质量标准的分析检测,也为栽培大黄代替野生大黄提供理论基础。

Objective To establish the HPLC fingerprints of Rheum tanguticum from Qinghai Region.Methods By RP-HPLC,acetonitrile and water including 0.04%H3P04 as mobile phase,gradient elution,flow rate was 1.0 mL/min,the detection wavelength was 280 nm,and the temperature was 40 C.Results The RSD of peak areas and retention time of common peak in the test of precision,repeatability,and stability has not reached 5%,respectively.Average similarity was 0.925 in R.tanguticum collected from different habitats.Conclusion All above results show that this method is simple,practicable,and reliable.It can be used as a standard analysis method in the quality control of R.tanguticum from Guoluo Region in Qinghai Province.It also provides a theory foundation on which the species cultivated could replace the wild ones.
