目的研究乳腺丸中阿魏酸在兔体内的药动学特性。方法采用高效液相色谱法测定阿魏酸的血药浓度。以香豆素为内标,甲醇-醋酸-水(38. 0:0. 5:61. 5)为流动相,ODS Hypers il C18柱(150 mm×4. 6 mm,5μm)为分析柱,检测波长为320 nm。结果阿魏酸在12. 5~1 600. 0μg/L呈线性,血浆最低检测质量浓度为6. 25μg/L,低、中、高3种质量浓度平均回收率分别为96. 17%、99. 80%、100. 23%,日内及日间RSD均<6%,5只兔单次ig乳腺丸后药-时曲线符合一室模型,药动学参数分别为AUC0-4(855. 29±100. 78)μg/(L. h),AUC0-∞(1 051. 99±125. 96)μg/(L. h),Cmax=(952. 71±119. 04)μg/(L. h),tmax=(0. 5±0. 00)h,t1/2=(1. 61±0. 21)h,Ke=(0. 46±0. 05)/h。结论本方法准确、重现性好,适用于乳腺丸中阿魏酸血药浓度测定及药动学研究。

Objective To study the pharmacokinetics of ferulie acid in rabbits in vivo after igadministering Ruxian Pill. Methods The concentrations of ferulic acid 1n plasma were determined by HPLC. Taking coumarin as internaI standard,a mixture of methanol-acetic acid-water(38. 0:0. 5:61. 5) was used as the mobile phase,and UV detection was performed at 320 nm. The analytical column was ODS Hypersii C18(150 mm×4. 6 mnl,5μm). Results The calibration curve was linear over the range ot 12. 5-1 600. 0μg/L and the measurable limit of plasma detection was 6. 25μg/L. The recovery was96. 1 7%,99. 80%,100. 23%. The RSD of the intra-day and the inter-days was 1ess than 6%. The plasma concentration-rime curve was consistent with one comparment model. The pharrnacokinetic paramerers were as follows:AUC0-4h(855. 29±100. 78)μg/(L·h),AUC0-∞(1 051. 99±125. 96)μg/(L·h),Cmax(952. 71±11 9. 04)μg/(L·h),tmax=(0. 5士0. 00)h,t1/2-(1. 6l±0. 21)h, Ke=(0. 46土O. 05)/h. Conclusion The method is accurate and precise with better reproducibility,which can be used for the determination of plasma level of ferulic acid and the study of its pharmacokineties for Ruxian Pill.
