[摘要]
目的观察仙芦抗癌胶囊的体内外抗肿瘤作用及对人肝癌HepG2细胞内Ca2+浓度([Ca2+]i)的影响,从而揭示仙芦抗癌胶囊的抗肿瘤作用机制。方法通过观察仙芦抗癌胶囊对S180A荷瘤小鼠瘤质量和H22小鼠生存时间的影响,观察其体内抗肿瘤作用。通过MTT法观察含药血清对肝癌HepG2细胞的细胞毒作用。通过Fluo-3/AM标记HepG2肝癌细胞,激光共聚焦扫描显微术测定肿瘤细胞[Ca2+]i,ATP酶试剂盒测定HepG2细胞膜Ca2+,Mg2+-ATP酶活性。结果仙芦抗癌胶囊(1.00、0.50、0.25g/kg)对S180A小鼠的瘤体生长有显著的抑制作用,对H22小鼠的生存时间有显著的延长作用。仙芦抗癌胶囊体外对HepG2细胞有细胞毒作用;对细胞[Ca2+]i有显著升高作用,高、中剂量(1.00、0.50g/kg)能够降低HepG2肝癌细胞膜Ca2+,Mg2+-ATP酶活性。结论仙芦抗癌胶囊有明显的体内外抗肿瘤作用,其作用机制为抑制肿瘤细胞膜Ca2+,Mg2+-ATP酶活性,增加细胞[Ca2+]i,从而诱导肿瘤细胞凋亡,达到抗肿瘤作用的目的。

objective The anthumor action both in vivo and in vitro,and the[Ca2+],of HepG2 cellswere studied tO show the antitumor mechanism of Xianlu Kang’ai Capsule.Methods The in viv0 antitu—mor action was studied by observing the variation of tumor weight and survival time of S1so A and H22 mice.The MTTassay was adopted to investigate the cellular cytotoxicity of the drug-serum On HepG2 cell.In—ternal Ca2+of HepG2 cell was mensurated by laser scanning confocal microscope(LSCM),and the activity Ca”,Mg”一ATPase of HepG2 eell membrance was determined by ATPase kit.Rcsults Xianlu Kang’ai Capsule(1.00,0.50,0.25 g/kg)inhibited the growth of cancer cells of S180 A mice and prolonged thesurvival time of H22 mice.Xianlu Kang’ai Capsule(1,00,0.50,0.25 g/kg)had strong cytotoxie effectson HepG2 cell and increased its internal Ca抖.The activity of Ca抖。M92+一ATPase WaS reduced by XianluKang’ai Capsule(1.00,0.50 g/kg).Conclusion Xianlu Kang’ai Capsule has antitumor effect both in vi—uD and in vitro.The mechanism is that Xianlu Kang’ai Capsule could increase the internaJ Ca2+of HepG2cell by reducing the activity of Ca“.Mg”一ATPase of HepG2 cell membrance to induce the apoptosis of tumot.
