目的采用电喷雾质谱技术研究附子的配伍规律,阐明八味地黄方与人参汤共服时产生毒性的化学物质基础。方法采用电喷雾质谱技术分别对八味地黄方加人参汤、附子加人参汤、附子加人参、附子加干姜、附子加甘草、附子加白术、次乌头碱对照品加白术共煎液中生物碱成分的相对质量分数及其变化规律进行研究。结果次乌头碱与白术、附子与白术、附子与人参汤、八味地黄方与人参汤共煎液中次乌头碱质量分数较高。结论八味地黄方与人参汤共煎液产生的毒性是由人参汤中的白术抑制了附子中次乌头碱等双酯型生物碱的水解,导致双酯型生物碱水平过高而引起的。甘草和干姜对附子具有解毒作用,人参亦不会导致共煎液的毒性增加。

Objective To investigate the compatible mechanism of Radix Aconiti Lateralis Preparata (RALP) by ESI-MS and clarify the chemical substances which could produce toxicity in the codecoction of Bawei Dihuang Recipe (BDR) and Ginseng Decoction (GSD). Methods The relevant alkaloids and their changing regularities in the codecoction of BDR+GSD, RALP+GSD, RALP+Radix Ginseng (RGS), RALP+Rhizoma Zingiberis (RZ), RALP+Radix Glycyrrhizae (RG), RALP+Rhizoma Atractylodis Macrocephalae (RAM), and hypaconitine+RAM were studied by ESI-MS. Results The relevant hypaconitine in the codecoction of hypaconitine+RAM, RALP+RAM, RALP+GSD, and (BDR+GSD) are more abundant. Conclusion The hydrolysis of diester-alkaloids, like hypaconitine, in RALP has been intensively restrained by RAM in GSD, which leads to the over high diester-alkaloids and induces the toxicity in the decoction of BDR and GSD. RG and RZ can repel the toxicity of RALP, and RGS does not make the toxicity of the codecoction increase.
