目的 研究复方861对人肝星状细胞(LX-2)中α-平滑肌肌动蛋白(α-SMA)基因表达的调节作用.方法应用基于SYBR-Green I的实时定量荧光PCR方法,研究复方861分别作用24、48、72 h后,LX-2细胞中α-SMA mRNA含量的变化情况.结果 复方861在48和72 h可以显著降低LX-2细胞中α-SMA mRNA含量.结论 复方861可抑制LX-2细胞中α-SMA基因的表达,提示其抗肝纤维化作用可能与其抑制LX-2细胞的活化有关.

Object To investigate the effects of Compound 861 (Cpd861)on expression of a-smooth muscle actin (a-SMA) in human hepatic stellate cells (LX-2 cells).Methods Real-time quantitative PCR method using SYBR Green I technology was taken to measure the effects of Cpd861 on expression of a-SMA mRNA in LX-2 cells treated by Cpd861 for 24,48 and 72 h.Results The expression levels of a-SMA mRNA decreased significantly in LX-2 cells exposed to Cpd861 for 48 h (P<0.05)or 72 h (P<0.01).Conclusion Cpd861 can reduce the expression levels of a-SMA mRNA in LX-2 cells.
