[摘要]
目的 研究白藜芦醇诱导胃癌细胞脱落凋亡的作用.方法 采用细胞形态学观察、Annxin-V和PI细胞染色和流式细胞仪检测,观察白藜芦醇(0.1、0.2、0.5 mmol/L)对脱落培养胃癌细胞B(3C823、SGC7901和HGC27的生长状态、细胞周期的影响及诱导脱落凋亡的作用.结果 倒置显微镜下发现脱落培养的胃癌细胞聚集成团生长,0.1 mmol/L白藜芦醇作用细胞后可抑制抗脱落凋亡的胃癌细胞聚集成团;流式细胞仪检测和Annxin-V和PI细胞染色表明0.5 mmol/L白藜芦醇可分别阻滞脱落培养的BG(2823、SGC7901和HGC27细胞于G0/G1、S、G2/M期,并可诱导这3株胃癌细胞发生脱落凋亡,其中自藜芦醇诱导HGC27细胞脱落凋亡比例达19.3%,远高于其他两株胃癌细胞.结论 白藜芦醇可诱导胃癌细胞发生脱落凋亡.

[Abstract]
Object To study the inducing of resveratrol on anoikis of gastric cancer cells.Methods Cell morphology observation,Annxin-V and PI staining,and Flow cytometry (FCM) assay were used to observe the inducing effect of resveratrol (0.1,0.2,0.5 mmol/L) on cell growth,cell cycles and anoikis of the gastric cancer cells (BGC823,SGC7901,and HGC27)cultured in suspension.Results Cluster growth of gastric cancer cells cultured in suspension was observed by invert microscope.Resveratrol (0.1mmol/L) could inhibit the gastric cancer cells resistant to anoikis to form cluster by FCM assay and Annxin-V and PI staining.Resveratrol (0.5 mmol/L) could arrest the suspended gastric cancer cells (BGC823,SGC7901,and HGC27)to G0/G1,S,G2/M phase respectively and induce the gastric cancer cells to anoikis,and the anoikis rate of HGC27 was around 19.3%,which was higher than that of other two cells.
