[摘要]
目的 研究以三七叶总皂苷制备人参皂苷C-K的酶转化工艺。方法 利用4种工业酶制剂对三七叶总皂苷进行转化,筛选出最佳酶制剂;以人参皂苷C-K的含量为指标,采用正交试验法进行酶转化工艺的优化;采用TLC及HPLC法检测酶转化产物。结果 酶转化三七叶皂苷的最佳工艺条件为20%β-葡聚糖苷酶,pH5.4,反应温度55℃,反应时间48h。反应时间的影响最大,其次是酶的体积分数。结论 β-葡聚糖苷酶对三七叶总皂苷的转化作用最强。

[Abstract]
Object To study the enzymatic translation of the saponins of Panax notog inseng leaves(SPNL)for preparation of ginsenoside compound K.Methods Four industrial enzymes were used to screen their ability of enzymatic translation of SPNL for preparation of ginsenoside compound K.Under the condition of orthogonal test,the affecting factors of enzymatic translation were studied and optim ized with the content of ginsenoside compound K,then analyzed by TLC and HPLC.Results The best optimal condition was obtained as follow ing:-0% forβ-glucanase concentration,pH 5.4,55 ℃ as reaction temperatur,48 h as the reaction time.The reaction time was the greatest affecting factors among the four ones,and enzyme concentration was the second.Conclusion β-Glucanase was the best of four industrial enzymes for this enzymatic translation technics.
