[摘要]
目的 研究蚕砂中提取、精制叶绿素类金属配合物 [Fe2+(Ⅰ)、Cu2+(Ⅱ)、Mn2+(Ⅲ)、Co2+(Ⅳ) ]作为抗活性氧 (O÷2 、H2O2 、OH· )模拟酶。方法 核黄素-蛋氨酸光照测其清除 O÷2 作用,H2O2 氧化维生素 C法测其催化H2O2 的分解,Fenton-苯甲酸钠荧光法测其对 OH·清除作用,小鼠肝匀浆法测其抗脂质过氧化作用。结果 1×10-5~ 1× 10 -6 mol/ L具有良好的清除 O÷2 作用,活性顺序为:Cu2+>Mn2+>Co2+>Fe2+;约 1× 10-7mol/ L具有分解 H2O2 作用,活性顺序为:Cu2+>Mn2+>Fe2+>Co2+;约 1× 10-8mol/ L 具有清除 OH·的功能,活性顺序为:Cu2+>Co2+>Fe2+>Mn2+;约 1× 10-8mol/ L 可使脂质过氧化产物明显减少,活性顺序为:Cu2+>Co2+>Mn2+>Fe2+。结论 蚕砂中叶绿素类金属配合物可作为抗多种活性氧 (O÷2 、H2O2、OH· )模拟酶

[Abstract]
Object Four metal chlorophylls produced by chlorophyll extracted from silkworm excrement were used as varied function mimicry of anti-active oxygen. Methods The first function, scavenging O÷2, has been proved by using riboflavine-methionine photoreduction methods. The second function, scaveging H2O2, has been demonstrated by using the oxidating Vc. The third function, scavenging OH·, has been demonstrated by using Fenton reaction. The complexes were measured by the liver homogenate technique of mice. Results Four model compounds could scavenge O÷2 in the concentration of 10-5 -10 -6 mol/L and the activity order was as follows: Cu2+ Mn2+ Co2+ Fe2+. All could decompose H2O2 in the concentration of 1×10-7 mol/L and the activity order was: Cu2+ Mn2+ Fe2+ Co2+. All could scavenge OH· in the concentration of 1×10-8 mol/L and the activity order was: Cu2+ Co2+ Fe2+ Mn2+. All showed that they had obvious action of decreasing the lipid peroxidation in the concentration of 1×10-8 mol/L. The activity order of anti-lipid peroxidation of complexes was as follows: Cu2+ Co2+ Mn2+ Fe2+. Conclusion All above-mentioned complexes were considered to be qualified mimic enzymes of anti-active oxygen.
