目的 研究抗癌活性蛋白火菇素的二级结构。方法 Fourier变换红外光谱和显微共焦拉曼光谱法。结果 火菇素蛋白的二级结构中含有较多的β折叠和α螺旋而无规则卷曲含量较少,“埋藏”和“暴露”的酪氨酸残基为34%和 6 6%,C-C-S-S-C-C-侧链为反式-扭曲-反式构型。结论 红外和拉曼光谱法可以准确地反映火菇素蛋白在溶液中的二级结构

[Abstract]
Object To determine the spatial structural properties of flammulin, an anti-tumor protein purified from the aqueous extract of basidiomes of Flammulina velutipes (Curf. ex Fr.) Sing. Methods The FTIR and Raman spectroscopic analysis methods were employed. Results It is found that protein of this sample contains predominately β-pleated sheet and α-helix. The majority of tyrosyl residues is exposed 66% and a few are buried 34%. This protein has trans-gauche-trans configuration of C-C-S-S-C-C linkage.Conclusion The FTIR and Raman spectroscopic analysis methods can be used to accurately determine the secondary structural properties of flammulin.
