[摘要]
目的 探讨当归多糖 (APS)对慢性粒细胞白血病细胞诱导生成树突状细胞 (DCs)的影响。方法 取慢性粒细胞白血病 (CML)患者骨髓单个核细胞,分别予粒 -巨噬集落刺激因子 (GM- CSF) /白介素-4 (IL-4)培养或GM- CSF/ IL- 4联合各浓度 APS(5 0,10 0,2 0 0 mg/ L)培养,普通光镜和电镜观察细胞形态,台盼蓝拒染法检测细胞成活率,流式细胞仪检测 DCs的免疫表型 (CD80,CD86,CD83),自体或异体混合淋巴细胞反应检测 DCs刺激 T淋巴细胞的增殖能力。结果 GM- GSF/ IL-4或 GM- CSF/ IL-4 / APS诱导培养的慢性粒细胞白血病骨髓单个核细胞均表现出典型的树突状形态而且表达高水平的免疫表型。GM- CSF/ IL- 4 / APS培养的 DCs的细胞成活率和增殖能力显著提高,CD83,CD80,CD86 表达显著升高,APS组慢性粒细胞白血病细胞诱导的树突状细胞 (CML-DCs)刺激 T淋巴细胞的增殖能力更强。结论 GM- CSF/ IL- 4 / APS培养的 DCs的 CD83、CD80 、CD86 表达率明显高于 GM-CSF/ IL-4组。 GM- CSF/ IL-4 / APS诱生的 CML-DCs刺激 T淋巴细胞增殖能力强于 GM- CSF/ IL-4组。 APS能促进 IL- 4和 GM- CSF对 CML- DCs的诱生与成熟。

Object To investigate the effect of A ngelica po1y saccharide (APS)on the induction of chronicmyelocytic leukan is ce1Ls into chronicmyelocytic leukaaniadendritic cells (CML-DCs). Methods Bonemarrow monocyts from CML patientsw ere cultured in GM-CSF/IL-4 or in GM-CSF/ IL-4 can bined with APS in each concentration(50,100,200 mg /L), respectively.The morphotype of CML-DCs was identified by opticalmicroscope or electronm icroscope, CM L-DCs viaility was calculated by rypan Blue exclusion. The phenotype of CML-DCs (CD80, CD86, and CD83) was identified by flow cytometry. The capability of stimulating auto-lym phocyteor allo-lym phocyte proliferat ion was tested withmixed leukocy tereaction (MLR). Results Bonem arrow monocy tes from CML patients, which were cultured in GM-CSF/IL-4 or in GM-CSF/IL-4/A PS showed typical morphotype and expressed the high level pheno type of CML -DCs. The capability of proliferation and the survival rate of CML-DCs were enhancedmarkedly and the expression of CD83, CD80, and CD86 on CML-DCs were significantly increased when CML-DCs were cultured in GM-CSF/IL-4/A PS.The capability of stimulating lymphocyte proliferation was more competentin 100mg/L A PS group. Conclusion The expression of CD83, CD80, and CD86 on CM L-DCs cultured in GM-CSF/ IL -4 /APS is sign if ican t ly h ighe r than tho se in GM -C SF /IL-4. The capability of CML-DCs of stimulating lymphocyte proliferation is more potentialin GM -CSF/IL-4 /A PS than in GM -C SF / IL-4. APS can promote the induction and mature of CML-DCs cultured in IL-4 and GM-CSF.
