目的 研究灯盏乙素拮抗硒对人肝细胞株 L- 0 2毒性作用。方法 采用噻唑蓝 (MTT)法研究灯盏乙素和硒对 L-0 2细胞生长的作用 ;倒置荧光显微镜观察细胞形态变化 ;测定丙二醛 (MDA )、巯基含量和总抗氧化能力等氧化还原相关生化指标。结果 0 .5 mm ol/ L 灯盏乙素与细胞作用 4 8h时达到促细胞生长最佳条件,且可以显著拮抗 1,5μm ol/ L硒对 L-0 2细胞生长的抑制作用 ;灯盏乙素和硒共同处理的细胞与仅用硒处理的细胞相比,细胞损伤程度明显减轻,MDA含量显著降低,巯基含量及体系抗氧化能力均显著增高。结论 灯盏乙素通过抗脂质过氧化和提高细胞抗氧化能力拮抗硒对 L-0 2细胞的毒性。

Object To investigate the antagonistic effect of scutellarinon the toxicity of selenium in human liver cells L-02. Methods MTT method was used to observe the effects of scutellarin and selenium on cell growth. The morphological changes of cells were observed by fluorescent microscope. The MDA level, thiolcontent, and total antioxidative capacity of cells were also measured. Results The optimum condition for scutellar in to promote cell growth was 0. 5 mmol/L in 48h culture period. Comparing with the only selenium treated group, 0.5 mmol/ L scutellarin significantly antagonized the inhibition of cell growth caused by 1 and 5 Lmol/ L selenium, suppressed the selenium-induced cell damage and the increase of MDA content, as well as increased the thiolcontent and cell antioxidation capacity. Conclusion Scutellarin markedly antagonizes the toxicity of selenium in human liver cells L-02 by inhibiting lipid perox idation and promoting cellular antioxidation capacity.
