[摘要]
目的 采用高效毛细管电泳法测定补阳还五汤及给药大鼠血清中阿魏酸的含量。方法 以毛细管区带电泳分离分析待测样品中的阿魏酸,内标法定量,紫外检测,缓冲液为 2 5 m mol/ L 硼砂 -甲醇 (85: 15 ),未涂层融硅毛细管 (5 2 cm× 5 0μm,有效长度 30 cm ),分离电压 2 0 k V,压力进样 3s,检测波长 32 0 nm,温度 30℃。结果 在上述条件下,阿魏酸在 6 min内得到很好的分离,血清中阿魏酸的加样回收率为 97.2 0 %,补阳还五汤中阿魏酸的回收率为 10 3.2 8%,RSD分别为 3.73%和 0 .91% (n=3)。结论 该方法可为血清样品及复方制剂中阿魏酸的含量测定提供参考。

[Abstract]
Object To develop a high-performance capillary electrophoresis method to determine ferulic acid concentrations in serum of rats treated with Buy ang Huanwu Decoction (BHD). Methods Capillary zone electrophoresisw as applied for ferulic acid assay, quantitative determination was based on internal standard and detection was carried on by direct UV. The electroly tebuf ferw as composed of 25mmol /L borax-methanol ( 85:15). Capillary electrophoresis was perform edusinga 52 cm (30cm to detector)×50μm fused-silica capillarytube. Separation voltage was 20 kV, sampling time was 3 s, detection wavelength was 320 nm, and the temperature was 30℃. Results Ferulic acid was successfully separated within 6min, the recoveries were 97.2% in serum and 103.28% in BHD, respectively. RSD were 3.73% and 0.91% (n=3), respective ly. Conclusion This method can supply reference for the determination of ferulic acid in serumsamples and BHD.
