[摘要]
目的 探讨白藜芦醇 (resveratrol,Res)对鼻咽癌细胞株 CNE- 2 Z增殖和凋亡的影响。方法 采用 MTT法检测 IC50 值,流式细胞术、Hoechst 332 5 8/PI荧光染色和 DNA琼脂糖凝胶电泳分析细胞凋亡。结果 不同浓度Res分别处理细胞 2 4,4 8和 72 h,其 IC50 值分别为 (10 9.2± 7.5 ),(83.6± 6.0 ),(5 4.3± 2.8) μmol/L,各 IC50 值间比较差异显著 (P<0.0 1)。2 5,5 0,10 0和 2 0 0 μm ol/L Res分别处理细胞 2 4 h后,5 0,10 0,2 0 0 μmol/L Res处理组经流式细胞术和荧光染色检测出的细胞凋亡率均明显高于对照组 (P<0.0 1),荧光染色可见典型的凋亡形态学改变,10 0,2 0 0 μmol/L Res处理组可见明显的 DNA梯带。结论 Res可通过诱导 CNE- 2 Z细胞株凋亡而抑制其增殖。

[Abstract]
Object To investigate the effects of resveratrol (Res) on the proliferation and apoptosis of nasopharyngeal carcinoma cell line CN E22Z. Methods IC50 values were detected by MTT assay. The apoptosis was analyzed by flow cytometry (FCM),Hoechst 33258/PI fluo rescence staining, and DNA agarosegel electrophoresis.Results After CN E-2Z cells were treated with different concent rations of Res for 24,48,and 72h, their IC50 values were (109.2±7.5),(83.6±6.0),and (54.3±2.8)μmol/L re-spectively (P<0.01). A f ter CN E-2Z cells were treated with 25,50,100,and 200 μmol/L Res for 24h,the apoptotic rates by both flow cytometry and fluo rescence staining at 50,100 and 200 μmol/L Res-treat-ed groups were sign ificantly higher than those in control groups (P< 0.01). The typical morphological changes in apoptosis were found by fluorescence staining. The results of DNA agarosegel electrophoresis showed that DNA ladder appeared obviously at 100 and 200 μmol/L Res-treated grou s. Conclusion Res can inhibit the proliferation of cellline CN E-2Z through the induction of apoptosis.
