[摘要]
目的 研究碱提山茱萸多糖的单糖组成及抗氧化活性。方法 经热水抽提后的山茱萸残渣用碱提取、盐酸中和、浓缩、透析、乙醇沉淀、DEAE-纤维素柱色谱分离,再通过 Sephadex G- 2 0 0柱色谱进一步纯化,得到白色粉末状多糖 PFCC 。结果 该多糖经过完全酸水解、薄层色谱、红外光谱分析,证明 PFCC 由木糖和葡萄糖以 18.8∶ 81.2摩尔比组成,平均相对分子质量为 7.5 7× 10 4。以羟基自由基 (· OH)清除剂苯甲酸和甘露醇为对照,用Fenton反应检测 PFCC 对· OH的清除作用,三者对· OH的清除率达 5 0 %所需浓度为 18,330,80μg/ m L。PFCC 可以有效抑制猪油和芝麻油的氧化,对 O÷2 的清除率达 5 0 %所需浓度为 35.0 μg/ m L。结论 该多糖为首次提取制得,具有良好的抗油脂氧化及清除自由基能力

Object To study the chemical composition and antioxidation activity of polysaccharide from the fruit of Cornus officinalis Zieb et Zucc methods The polysaccharide PFCC Ⅰ was isolated from C officinalis through NaOH extraction, neutral reaction, ethanol precipitation and DEAE cellulose column chromatography It was further purified by Sephadex G 200 column chromatography Results The molecular weight of PFCC Ⅰ was 7 57×10 4 Monosaccharide composition analysis by TLC and HPLC showed that it was composed of xylose and glucose in a molar ratio of 18 8∶81 2 PFCC Ⅰ had strong activities of oxidation resistance to axunge and gingili It also had an action to scavenging hydroxyl free radical (·OH) generated by Fenton raction and superoxide anion free radical (O ÷ 2) generated by pyrogallol autoxidation system The scavenging activity to ·OH and O ÷ 2 was represented by the content scavenging 50% of all radicals or anion (Ec 50 ), which were 80 μg/mL and 35 0 μg/mL respectively Conclusion This polysaccharide is isolated from C officinalis for the first time, which could resist oxidation of axunge and gingili and scavenge free radical effectively
