目的 通过离体实验观察人肠道内细菌对七叶皂苷的代谢。方法 离体培养人肠道内细菌,与七叶皂苷厌氧温孵,采用硅胶柱色谱和制备型高效液相色谱等方法对七叶皂苷的肠菌代谢产物进行分离和代谢转化产物的鉴定。结果 代谢产物经 IR、MS和 NMR鉴定为 :2 1β-巴豆酰基原七叶皂苷元 ( ),2 1β-当归酰基原七叶皂苷元( ),2 1β-巴豆酰基 - 2 2α-乙酰基原七叶皂苷元 ( ),2 1β-当归酰基 - 2 2α-乙酰基原七叶皂苷元 ( )。其中 和 为首次发现的新天然产物。结论 在离体条件下,七叶皂苷可被人肠内菌代谢为 2 1β-巴豆酰基原七叶皂苷元 ( )、2 1β-当归酰基原七叶皂苷元 ( ),2 1β-巴豆酰 - 2 2α-乙酰基原七叶皂苷元 ( )、2 1β-当归酰基 - 2 2α-乙酰基原七叶皂苷元( )。

[Abstract]
Object To study the aescin metabolism in human intestinal flora in vitro methods Human intestinal flora and aescin were incubated in vitro TLC and HPLC were used to examine aescin and its metabolites; the metabolites were separated and purified by silica gels column chromatography and pre paration high performance liquid chromatography Results Their structures were identified by IR, MS and NMR as 21β tigloyl protoaescigenin (Ⅰ), 21β angeloyl protoaescigenin (Ⅱ), 21β tigloyl 22α acetylprotoaescigenin (Ⅲ), 21β angeloyl 22α acetylprotoaescigenin (Ⅳ) Among them Ⅱ and Ⅳ were new natural products found from this plant. Conclusion Aescin can be in vitro metabolized by intestinal flora into 21β tigloyl protoaescigenin (Ⅰ), 21β angeloyl protoaescigenin (Ⅱ), 21β tigloyl 22α acetylprotoaescigenin (Ⅲ), 21β angeloyl 22α acetylprotoaescigenin (Ⅳ)
