[摘要]
目的 对以往建立的人血清中阿魏酸浓度测定的HPLC法加以改进,并测定了血瘀证患者口服川芎丹参煎剂后阿魏酸的血药浓度。方法 以香豆素为内标,甲醇-水-醋酸(40∶59.7∶0.3)为流动相,Diamonsil(TM)色谱柱(150mm×4.6mm,5μm),流速1.0mL/min,以二极管阵列检测器测定322nm波长下的数据,以沸水浴法加甲醇处理血清样品。结果 阿魏酸和内标分离完全,阿魏酸在32.18~965.38ng/mL范围内线性关系良好(r=0.9981),日内和日间RSD<10%,平均回收率为(99.3±5.4)%,最低检测限为1.6ng/mL,最低检测浓度为15.7ng/mL。结论 此法简单、快速、灵敏而准确,适用于人血清中阿魏酸血药浓度的测定。

Object To improve the HPLC method for deter mination of ferulicacid(FA) and to quantitateserum FA concentration in patients with blood stasis after oraladmini stration of decoction of Ligusticum chuan xiong Hort.and Salviavemiltiorrhizae Bunge root. Methods The serumsamplew astreated with aboiling water bath.The coumarin was used asinternal standard compound. The mixed solvent consisted of methanol, water and aceticacid(40∶59.7∶0.3) was used as elutriant.The flow rate was 1.0 mL/min.The Diam on sil TM column(150mm×4.6mm,5μm)was used.FA was detected at UV -322nm and the peak of FA in serum was recognized by photodiode- array detector . Results FA and in ternal standard compound were separated completely under the conditionde scribed as above.F A showed agoodline arity in the range of 32.18~965.38ng /mL(r=0.9981). The R S D was less then 10%, and the minimum detection amount was 1.6ng/m L and the minimum detection concentration sinseum was 15.7ng/ mL. Conclusion Improved HPLC method is simple,sensitive,rapid and accurate.It can be applied to detect FA concent ration in humanserum.
