用正交法研究了不同条件对灵芝菌生长影响, 从中选出了灵芝菌液体深层发酵的优化培养基:葡萄糖3.6%, 蛋白胨0.4%, pH6.0, 酵母膏0.2%, KH2PO40.1%, MgSO4·7H2O0.05%, Vit B10.005%, 每升发酵液产干菌丝体15.6g, 粗多糖0.72g.经葡聚糖凝胶层析对灵芝胞外多糖进行了分离纯化, 共有5个组分, 并用红外光谱、紫外光谱, 气相色谱等手段进一步分析了灵芝胞外多糖的组成, 结果表明灵芝胞外多糖系由甘露糖、果糖、葡萄糖通过糖β-D-糖苷键构成.

Optimal medium for deep layer fermentation of Ganoderma lucidum (Leyss. ex Fr. ) Karst was selected and conditions for the isolation and purification of its ex tracellular polysaccharide studied for the purpose to develop a new source for t he production of this valuable TCM. Factors influencing the growth of G. lucidum on different media were studied by orthogonal experimental de sign and polysaccharide isolated and purified by Sephadex gel filtration. The s tr uctural composition was analysed by UV, IR and GC chromatography. The best medi um for the fermentation was found to be composed of glucose 3.6%, peptone 0.4%, y east extract 0.2%, KH2PO4 0.1%, MgSO4·7H2O 0.5% and VB1 0.005%, at a pH va lue of 6.0. 15.6 g dry mycellia and 0.72 g crude polysaccharide per liter ca n be ob tained from the fermentation broth. The polysaccharide was composed of glucose, Dfructose and mannose. The method needs further scale up study. 【Key Words】: Ganoderma lucidum (Leyss. ex Fr.)
