采用气管条浴槽实验方法,观察青心酮(DHAP)对离体豚鼠气管螺旋条张力的影响。结果发现:DHAP明显松弛豚鼠的离体气管平滑肌 ;抑制由乙酰胆碱(Ach)、组胺(His)和氯化钾所致离体豚鼠气管平滑肌的收缩反应;对5-羟色胺(5-HT)诱发的由内钙释放引起的|相收缩和外钙内流所致的|相气管平滑肌收缩均有一定的抑制作用。3.96× 10-5 m ol/L DHAP则仅抑制|相收缩。随着其剂量的增大(>10-4 mol/L),相收缩也被抑制,上述抑制作用均呈剂量依赖性;消炎痛对 DHAP松驰离体气管平滑肌作用无明显影响。提示 DHAP这一效应可能与其钙拮抗有关,而与其抑制环加氧酶的作用关系则不甚密切。

The Effects of 3,4-dihydroxyacetophenone (DHAP) o n the tension of isolated guinea-pig trachea spirals were studied by tension re cording method. It was found that DHAP relaxed the basal tension and markedly d e pressed the contractile response induced by the spasmogens (Ach, histamin, KCl, 5-HT) in a concentration-dependent manner. But indomethacin, a cyclooxygenase inhibitor, showed no influence on the relaxant effect of DHAP against the contra ction of the spirals caused by Ach. DHAP could abolish the contraction induced b y 5-HT both in phase 1 (release of intracellular calcium) and phase 2 (entrance of extracellular calcium). These results suggested that the relaxant effect of DHAP on isolated guinea-pig trachea spirals may be resulted from a decreased co ncentration of intracellular calcium.
