[摘要]
建立了测定离体透皮接受液中丹参酮ⅡA的HPLC测定法。流动相:甲醇-水(85:15);紫外检测波长254nm,流速1mL/min。实验表明在230~2300ng/mL浓度范围内,丹参酮Ⅱ_A的浓度与峰高的线性关系良好,r=0.9984。皮肤渗透液中的丹参酮Ⅱ_A的平均回收率为101.62%,日内、日间 RSD分别4.45%及4.23%。该法简便,分离完全,结果可靠,适用于丹参酮 Ⅱ_A的离体透皮液浓度的测定。

HPLC method was established for the determination of Tanshinone ⅡA(Tan ⅡA) concen-tration in the Valia-Chien diffusion cell. The mobile phase consisted of methanol:water (85:15). The detected wave length was 254 nm and flow rate was 1 ml/min. The assay linearity was determined over the range of 230-2300 ng/ml in 5% PEg 600 permeability solution (r=0. 9984). The mean recovery of Tan ⅡAwas 101.62 0 o. The within day and day to dayv ariati ons were 4.45% and 4.23% respectively which were both less than 10%The method is simple,rapid, and reliable. It is suitable for the determination of Tan.ⅡAconcentration.
