[摘要]
目的 研究方枝守宫木Sauropus quadrangularis中的黄酮类化学成分及其抗氧化活性。方法 综合应用正相硅胶柱色谱、Sephadex LH-20凝胶柱色谱、高压制备液相色谱以及重结晶等方法进行分离纯化,通过核磁共振波谱以及质谱等技术鉴定化合物的结构,采用DPPH法评价化合物的体外抗氧化活性。结果 从方枝守宫木地上部分的乙醇提取物中分离得到23个黄酮类化合物,分别鉴定为山柰酚(1)、槲皮素(2)、儿茶素(3)、表儿茶素(4)、山柰酚-3-O-β-D-木糖苷(5)、山奈酚-3-O-(6″-O-乙酰基)-β-D-葡萄糖苷(6)、corbulain Ib(7)、山柰酚-3-O-α-L-鼠李糖苷(8)、槲皮素-3-(2''-β-D-葡萄糖基)-α-L-鼠李糖苷(9)、槲皮素-3-O-(2''-O-乙酰基)-α-L-鼠李糖苷(10)、槲皮素-3-O-(3''-O-乙酰基)-α-L-鼠李糖苷(11)、山柰酚-3-O-(5″-O-乙酰基)-α-呋喃阿拉伯糖苷(12)、槲皮素-3-O-(5″-O-乙酰基)-α-呋喃阿拉伯糖苷(13)、槲皮素-3-O-α-呋喃阿拉伯糖苷(14)、山柰酚-3-O-α-L-呋喃阿拉伯糖苷(15)、槲皮素-3-O-β-D-木糖苷(16)、槲皮素-3-O-α-L-鼠李糖苷(17)、8-(2-吡咯烷酮-5-基)-(−)-表儿茶素(18)、山柰酚-3-O-芸香糖苷(19)、芦丁(20)、山柰酚-3-O-β-D-葡萄糖苷(21)、莽吉柿酮D(22)、槲皮素-3-O-β-D-葡萄糖苷(23),其中化合物10~11、13、17具有较好的DPPH自由基清除活性,清除率达到75%以上。结论 所有化合物均为首次从方枝守宫木中分离得到,化合物10~11、13、17具有较好的抗氧化活性。

[Abstract]
Objective To study the flavonoids from Sauropus quadrangularis and their antioxidant activities. Methods The constituents were isolated from 95% ethanol extract of the aerial part of S. quadrangularis and purified by silica gels column chromatography, Sephadex LH-20 column chromatography, high pressure preparative HPLC and recrystallization method. Their structures were identified based on NMR, MS and other spectroscopic techniques. Their antioxidant activities were evaluated by scavenging capacity on DPPH. Results A total of 23 flavonoids were isolated from S. quadrangularis and identified as kaempferol (1), quercetin (2), (+)-catechin (3), epicatechin (4), kaempferol-3-O-β-D-xylopyranoside (5), kaempferol-3-O-(6″-O-acetyl)-β-D-glucopyranoside (6), corbulainIb (7), kaempferol-3-O-α-L-rhamnoside (8), quercetin-3-(2''-β-D-glucosyl)-α-L-rhamnoside (9), quercetin-3-O-(2″-O-acetyl)-α-L-rhamnoside (10), quercetin-3-O- (3″-O-acetyl)-α-L-rhamnoside (11), kaempferol-3-O-(5″-O-acetyl)-α-arabinofuranoside (12), quercetin-3-O-(5″-O-acetyl)-α-arabinofuranoside (13), quercetin-3-O-α-arabinofuranoside (14), kaempferol-3-O-α-L-arabinofuranoside (15), quercetin-3-O-β-D-xylpyranoside (16), quercetin-3-O-α-L-rhamnoside (17), 8-(2-pyrrolidinone-5-yl)-(−)-epicatechin (18), kaempferol-3-O-rutinoside (19), rutin (20), kaempferol-3-O-β-D-glucopyranoside (21), garcimangosone D (22), quercetin-3-O-β-D-glucoside (23). Compounds 10—11, 13, 17 had the stronger DPPH scavenging abilities and the clearance rate exceeded 75%. Conclusion All compounds are isolated from S. quadrangularis for the first time. Compounds 10—11, 13, 17 possess good antioxidant activities.
