目的 研究红花Carthamus tinctorius的化学成分。方法 采用大孔吸附树脂、硅胶、Sephadex LH-20、ODS柱色谱以及半制备型HPLC等分离技术对红花化学成分分离纯化,运用理化性质和UV、IR、HR-ESI-MS、NMR、ECD等谱学数据鉴定化合物结构。采用氧糖剥夺/复氧(OGD/R)损伤的H9C2心肌细胞模型,对分离得到的化合物进行心肌损伤保护活性筛选。结果 从红花95%乙醇提取物大孔树脂95%乙醇洗脱部位中共分离鉴定了4个化合物,分别为 (−)-(3Z,1R,5S,9S,11R)-3-烯-石竹-8(13)-烯-5-醇-14-O-β-D-吡喃葡萄糖苷(1)、(−)-(3Z,1R,5S,9S)-3-烯-石竹-8(13)-烯-5-O-β-D-吡喃葡萄糖苷(2)、(−)-桃金娘烯醇-10-O-β-D-吡喃葡萄糖苷(3)、香叶醇-1-O-β-D-吡喃葡萄糖苷(4)。化合物1和4在25 μmol/L浓度下分别对OGD/R损伤的H9C2细胞存活率提高了(25.01±2.01)%和(29.99±0.37)%。结论 化合物1和2为新的倍半萜苷类化合物,命名为红花萜苷A(1)和红花萜苷B(2);化合物3和4为首次从红花属植物中分离得到的单萜苷类化合物,体外活性测试表明化合物1和4具有良好的心肌损伤保护活性。

[Abstract]
Objective To investigate the chemical constituents of Carthamus tinctorius.Methods The compounds were isolated and purified by various column chromatography over microporous resin, silica gel, Sephadex LH-20, ODS, and semi-preparative HPLC. The structures were elucidated by physicochemical characteristics and spectroscopic data including UV, IR, HR-ESI-MS, NMR, as well as ECD calculations. All the compounds were evaluated for their protective effects against OGD/R-stimulated H9C2 cell injury. Results A total of four compounds were isolated from the 95% EtOH extract of C. tinctorius and elucidated as (−)-(3Z,1R,5S,9S,11R)-3-en-caryophyll-8(13)-en-5-ol-14-O-β-D-glucopyranoside (1), (−)-(3Z,1R,5S,9S)-3-en-caryophyll-8(13)-en-5-O-β-D-glucopyranoside (2), myrtenyl-10-O-β-D-glucopyranoside (3), geranyl-1-O-β-D-glucopyranoside (4). Compounds 1 and 4 could significantly protect OGD/R-induced H9C2 cell injury, with increase of cell viabilities by (25.01 ±2.01)% and (29.99 ±0.37)% at the concentration of 25 μmol/L, respectively. Conclusion Compounds 1 and 2 were new sesquiterpene glucosides, named saffloterpenoside A (1) and saffloterpenoside B (2). Compounds 3 and 4 were isolated from the Carthamus plants for the first time. Compounds 1 and 4 exhibited protective effects on OGD/R-injured H9C2 cells.
