[摘要]
目的 建立基于多成分含量和多元统计分析的提取工艺优化方法,并优化益艾康胶囊水提取工艺。方法 以正交试验法,选择L9(34)正交表考察煎煮次数(A)、煎煮时间(B)、加水量(C)3个因素的不同水平对益艾康胶囊水提工艺的影响;以HPLC法测定样品中甘草苷、甘草酸铵、黄芩苷、芍药苷、黄芪甲苷和毛蕊异黄酮葡萄糖苷的含量并进行方法学研究;通过主成分分析法对实验数据统计分析,以主成分得分为因变量对正交试验结果进行方差分析,考察各影响因素对提取效果的影响;以多重比较法优选各因素水平。结果 优选得到的提取工艺为加8倍量水,煎煮3次,每次煎煮1 h。结论 所建立的研究方法既能充分反映样品内在质量,又能够简化数据结构,可为中药复方提取工艺研究提供参考。优选得到的工艺简便易行,可作为益艾康胶囊水提取工艺。

[Abstract]
Objective To establish an extraction process optimization method based on multi-component content determination and multivariate statistical analysis, and optimize the water extraction process factors of Yiaikang Capsules (YAKC). Methods L9(34) orthogonal table was selected to investigate the effects of different levels of extracting times (A), extracting time (B) and water addition (C) on water extraction process of YAKC. The contents of liquiritin, ammonium glycyrrhizinate, baicalin, paeoniflorin, astragaloside IV, and calycosin-7-O-β-D-glucopyranoside in the orthogonal samples were determined by HPLC, and related methodological studies were carried out. The experimental data was treated by principal component analysis, principal component score was calculated and chosen as dependent variable, variance was analyzed to evaluate the influence of various factors. The optimum levels of extraction factors were selected by multiple comparison. Results The optimized extraction process method was as following:Eight times of water be added, the drug be extracted for three times, each time for 1 h. Conclusion The established method can reflect the intrinsic quality of the sample and simplify the data structure, which can provide reference for the further Chinese medicine compound extraction process study. The optimized extraction method of YAKC is simple and reliable.
