[摘要]
目的建立复方五仁醇胶囊含药血清中五味子醇甲、五味子醇乙、五味子甲素和五味子乙素的测定方法。方法采用高效液相色谱法。色谱条件:Lichrosphere C18色谱柱(250 mm×4.6 mm,5μm),Phenomenex De-scription C18保护柱(4.0 mm×3.0 mm);
水-乙腈梯度洗脱;
体积流量1.0 mL/min;
柱温30℃;
检测波长210 nm。结果五味子醇甲、五味子醇乙、五味子甲素和五味子乙素分别在0.051 2~0.768 0μg(r=0.999 5)、0.054 0~0.810 0μg(r=0.999 6)、0.012 3~0.184 5μg(r=0.999 8)、0.039 8~0.597 0μg(r=0.999 6)线性关系良好,含药血清中4种成分的平均质量浓度分别为8.021 1、6.231 0、0.530 8、5.851 0μg/mL。结论该法简便、准确、专属性强,可用于复方五仁醇胶囊含药血清中木脂素类成分的测定。

[Abstract]
Objective To determine schisandrin,schisandrol B,deoxyschizandrin,and schisandrin B in serum containing drug of Compound Wurenchun Capsula.Methods An HPLC method was set up.Li-chrosphere C18 column(250 mm ×4.6 mm,5 μm) and Phenomenex Description C18(4.0 mm×3.0 mm)protective column were used.Acetonitrile-water was used as gradient mobile phase.The flow rate was 1.0 mL/min.The column temperature was 30 ℃ and the detection wavelength was 210 nm.Results The linear ranges of schisandrin,schisandrol B,deoxyschizandrin,and schisandrin B were within 0.051 2-0.768 0 μg(r=0.999 5),0.054 0-0.810 0 μg(r=0.999 6),0.012 3-0.184 5 μg(r=0.999 8),and 0.039 8-0.597 0 μg(r=0.999 6),respectively.The average concentration of these four lignans in serum containing drug were 8.021 1,6.231 0,0.530 8,and 5.851 0 μg/mL,respectively.Conclusion This method is easy,sensitive,specific,and accurate for the assaying of the four lignans in serum containing drug of Compound Wurenchun Capsula.
