目的: 研究朝鲜淫羊藿Epimedium koreanum中具有抗骨质疏松活性的成分, 考察化合物对大鼠骨肉瘤细胞UMR106的增殖和分化的影响。方法: 以大鼠骨肉瘤细胞UMR106的增殖作为活性追踪指标, 运用各种现代分离手段对朝鲜淫羊藿的水提取物进行活性成分的追踪分离, 用光谱学方法对得到的化合物进行结构鉴定, 同时检测化合物的活性。结果: 分离得到5个木脂素类化合物, 分别鉴定为: 9′-(α-吡喃鼠李糖基)-3, 5′-二甲氧基-3′:7, 4′:8-二环氧新木脂素-4, 9-二醇(Ⅰ)、柏木苷A(Ⅱ)、(+)-环合橄榄树脂素(Ⅲ)、(+)-南烛木树脂酚(Ⅳ)、(+)-异落叶松树脂醇(Ⅴ)。检测了化合物Ⅰ~Ⅴ对UMR106细胞增殖和分化的影响。结论: 化合物Ⅰ为新化合物, 命名为柏木苷C; 化合物Ⅱ、Ⅳ、Ⅴ均为首次从该属植物中分离得到。化合物Ⅱ~Ⅴ对UMR106细胞的增殖及碱性磷酸酶的活性都呈现一定程度的促进作用, 化合物Ⅰ只促进细胞的增殖。

Objective: To study the antiosteoporotic constituents from Epimedium koreanum and the effects of isolated compounds on proliferation and differentiation of the osteoblasts-like UMR l06 cells.Methods: Guided by the bioactivity on osteoblasts-like UMR l06 cells, the constituents in water extract of E.koreanum were separated with chromatographic Methods: The structures of the isolated compounds were identified by spectral analyses, while their effects on the proliferation and alkaline phosphatase (ALP)activities of UMR 106 cells were observed.Results: Five lignans were obtained, they are 9'-(a-rhamnopyranosyl)-3, 51_dimethoxy-37:7, 47:8-diepoxyneolignan-4, 9-diol(I), cupressoside A(I), (+)-cycloolivil(11), (+)-lyoniresinol(Ⅳ), and(+)-isolariciresinol(V).The effects of compound I-V on proliferation and ALP activities were determined.Conclusion: Compound I is a new compound, named cupressoside C; 1, Ⅳ, and V were isolated from E. koreanum for the first time. Compound l-V show stimulating effects on both proliferation and ALP activites of UMR l06 eell.while compound I could only stimulate the proliferation.
