[摘要]
目的:观察积雪草苷对脂多糖(LPS)诱导急性肺损伤(ALI)小鼠的保护效应。方法:56只Balb/c小鼠随机分为对照、假手术、模型、溶媒、积雪草苷(5、15、45 mg/kg)7组, 每组8只。LPS(2.5 mg/kg)气管滴注造ALI模型, 24 h后HE染色观察小鼠肺组织病理改变, 计算肺湿质量/干质量值;收集支气管肺泡灌洗液(BALF)后行白细胞计数和分类, BCA法检测其蛋白的量;化学法检测肺组织髓过氧化物酶(MPO)活性。结果:积雪草苷各剂量组呈剂量依赖性减轻LPS所致ALI模型肺组织病变情况, 减轻肺水肿, 减轻中性粒细胞的浸润, 抑制蛋白渗出。抑制肺组织中MPO活性。结论:积雪草苷对LPS诱导ALI有保护效应。

[Abstract]
Objective: To observe the protective effect of asiaticoside(AS)on acute lung injury(ALI) of mice induced by lipopo1ysaccharide(LPS). Methods: Balb/c mice(56)were randomlydivided into control, Shamoperation, model, model+vehicle, andmodel+asiatieoside(5, 15, and 45mg/kg)groups. The model of ALl was established by intratraeheal instillation of LPS 20 uL(2. 5 mg/kg). Mice were killed 24 hlater, pathological changes of lung were observed with HEstaining, the ratio of pulmonarywet weight to dry weight(W/D) of the lungwere measured, the number and classification of leucocyte of bronchoalveolar lavage fluid(BALF), were counted, the protein of BALF was detected by BCA, the activity of my eloperoxidase(MPO)in lung was detected by MPO kit. Results: Asiaticoside could alleviate the damage of lung caused by LPS through decreasing alveolar interstitial tissue edema, leucocyte infiltrationin. and reducing the activity of MPO of lung in a dose-dependent manner. Conclusion: Asiaticoside has the protective effects against ALI of mice indueed by LPS.
