[摘要]
目的:分析黄芩苷对小鼠T淋巴细胞体外活化的影响, 探讨其对免疫系统的作用, 阐明其免疫抑制作用机制。方法:无菌分离小鼠淋巴结, 制备淋巴细胞悬液, 以不同终质量浓度的黄芩苷预孵育淋巴细胞4 h, 再以刀豆蛋白A(Con A)或佛波醇酯类多克隆刺激剂(PDB)诱导小鼠淋巴细胞活化, 利用流式细胞术分别结合双色免疫荧光单克隆抗体染色技术检测CD3+T细胞早期活化标志CD69和中期活化标志CD25分子的表达。结果:黄芩苷(10、20、30 mg/L)能明显抑制Con A或PDB诱导T细胞活化分子CD69和CD25的表达, 且呈浓度依赖性。结论:黄芩苷能够明显抑制多克隆刺激剂Con A或PDB诱导的T细胞早期和中期活化, 且呈明显的量效关系。提示黄芩苷在T细胞活化过程的信号转导中有抑制作用, 从而可以通过抑制T淋巴细胞的活化, 发挥免疫抑制作用, 是-种潜在的免疫抑制剂。

0bjective: To investigate the effect of baicalin on the activation of the mouse Tlymphocytes in vitro. in order to elucidate the mechanism of the immunosuppressive effect of baicalin. Methods: To isolatesterilely lymphocytes suspensionfrom mouselymph nodes, prepare lymphocytes which were pretreated with different concentration of baicalin for 4 h and stimulated with polyclonal activators Con A or PDB, fluorescence conjugated monoclonal antibodies and flowcycometry were used to detect the expression of CD69 and CD25 of mouse CD3+ Tlymphocytes activated. Results: Baicalin(10, 20, and 30mg/L)could inhibit the expression of CD69 and CD25 of Tlymphocytes activated by Con A or PDB in a dose-dependent manner. Conclusion: Baicalin could inhibit the initial-period and mid-period activation of mouse Tlymphocytes stimulated by Con A or PDB, which suggests that baicalin have the inhibitory effect on signaling pathway of Tlymphocytes activation and be a potential effective immunoinhibitory agent.
