从采集于四川省木里县的云南红豆杉Taxusyunnanensis的树皮中分得一个具有较强生物活性的新紫杉烷类似物,命名为紫杉次碱(taxoline,Ⅰ)。测定其化学结构通过解析其1HNMR、13CNMR、FABMS、DEPT谱及1H-1HCOSY二维谱与化学方法。Ⅰ经本所药理室用1H-TDR掺入法测定,对P388淋巴细胞白血病DNA合成的抑制强度比作为对照品的紫杉醇大6.5倍(Ⅰ的IC50为1.12μg/mL,紫杉醇为7.4μg/mL)。进一步的抗癌活性正在评价中。

A novel taxane,with more potent bioactivity,was isolated from Taxus yunnanensis,collect ed in Molin county,Sichuan Province.It is named taxoline (Ⅰ). The chemical structure was determined by 1HNMR, 13CNMR,FABMS,DEPT, 1H-1HCOSY spectra and chemical methods. Its inhibition of P388 leukaemic lymphocyte was 6. 5 times that paclitaxel (IC50 1. 2μg/mL forⅠ;7. 4 μg/mL for paclitaxel). Further estima tion of antitumor activity for Ⅰ is ongoing.
