[摘要]
目的 以黄连为对象探讨中药谱效关系研究新方法。方法 建立优化基于ATP生物发光的微量抑菌检测方法,与微量稀释法抑菌结果进行相关性考察。以所建立的抑菌检测方法检测黄连抑菌活性,整合黄连的化学与抑菌活性图谱,探讨黄连化学组分与活性之间的关系。结果 所建立的基于ATP生物发光抑菌检测法与微量稀释法相关性良好,分别检测大肠杆菌和金黄色葡萄球菌所得相关系数r值分别为0.998和0.957。黄连谱效相关研究结果显示,黄连中除主要有效成分黄连素外,还可能含有微量高效抑菌成分。结论 基于ATP生物发光的抑菌活性检测方法适用于中药抑菌活性的谱效关系研究,为中药抑菌活性谱效相关研究提供新的技术手段。

Objective Take Rhizoma Coptidis as the object to explore new methods of Chromatogram-Effect Relationship of traditional Chinese medicine. Methods Establish and optimize the trace bacteriostatic detection method based on ATP bioluminescence, and investigate the correlation of the results between ATP bioluminescence and microdilution method. Detect the antibacterial activity of Rhizoma Coptidis by Established antibacterial detection method. Integrate the chemical and activity profile of Rhizoma Coptidis to discuss the relationship between the chemical composition and the activity. Results The results obtained by Established ATP bioluminescence assay correlate well with that by microdilution method, the r value of Escherichia coli and Staphylococcus aureus were 0.998 and 0.957, respectively. The integration activity spectra showed that except berberine there also may be exist high antibacterial activity substances in Rhizoma Coptidis. Conclusion ATP bioluminescence can be used in the study of Antibacterial Chromatogram-Effect Relationship of traditional Chinese medicine. Provide a new method for the quality standards of traditional Chinese medicine.
