[摘要]
目的 建立山楂中化学成分的超高液相/电喷雾–飞行时间/质谱(UPLC/ ESI-TOF/MS)的分析方法。方法 Acquity UPLC BEH C18色谱柱(100 mm×2.1 mm,1.7 μm),流动相0.1%甲酸水溶液–乙腈,梯度洗脱;体积流量0.3 mL/min,柱温50 ℃,检测波长范围260~360 nm,进样量1 μL。ESI离子源,飞行时间质谱检测器,对比对照品进行鉴别。结果 共鉴定出13个化合物,分别为苹果酸(1)、枸橼酸(2)、绿原酸(3)、花青素B2(4)、表儿茶素(5)、花青素B3(6)、牡荆素葡萄糖苷(7)、芦丁(8)、金丝桃苷(9)、异槲皮苷(10)、槲皮素(11)、熊果酸(12)、熊果醇(13)。结论 建立了一种简单、快速的UPLC/ESI-TOF/MS方法对山楂中化学成分进行了鉴定。

[Abstract]
Objective To identify the chemical constituents of Crataegus pinnatifida var. major N. E. Brown by UPLC/ESI-TOF/MS. Methods The method of identification was performed on Acquity UPLC BEH C18 column (100 mm×2.1 mm, 1.7 μm), the mobile phase consisted of 0.1% formic acid aqueous solution and acetonitrile with gradient elution program. The injection volume was 1 μL at the flow rate of 0.3 mL/min. The column temperature was 50 ℃, and the detection wavelength was set from 260 to 360 nm. The detector was TOF Mass Spectrometer with an ESI ion source. The data were compared with those of the reference substances. Results Thirteen compounds were identified as malic acid (1), citric acid (2), chlorogenic acid (3), anthocyanin B2 (4), epicatechin (5), anthocyanin B3 (6), vitexin-glucoside (7), rutin (8), hyperoside (9), isoquercetin (10), quercetin (11), ursolic acid (12), and uvaol (13). Conclusion The established method for the determination of chemical constituents of C. pinnatifida var. major using UPLC/ESI- TOF/MS is simple and reliable.
